基于Cytb基因序列对入境集装箱中截获的疑似鼠样本的种类鉴定

  摘要:目的 根据线粒体Cytb基因部分序列,对从入境集装箱中截获的疑似鼠样本进行鉴定种类。方法提取样本中的DNA,采用PCR方法进行线粒体Cytb基因扩增,对目的片段测序,通过blast比对进行同源性比对。从Genbank获取宁波口岸常见8个鼠种24个样本的Cytb基因序列作为参照,利用生物信息学软件Mega5.0进行系统进化分析。结果样本DXR1110与小家鼠同源性最高,DXR1203与褐家鼠同源性最高,系统进化树显示不同鼠种间形成独立分支。结论Cytb基因分析是进行鼠种鉴定的有效手段。

  Species indentification of suspected samples of rats found inimported containers based on Cytb gene

  Abstract:   Objective   To identify species of twosuspected samples of rats which found in imported containers basedon partial sequences of mitochondrion Cytb gene.  Methods  Extract DNA from samples, the partial sequences of Cytb genewere amplified using PCR. The target segments were sequenced andsequences alignment had been done through blast. In order to buildthe phylogenetic tree, 24 cytb sequences were downloaded fromGenbank. These sequences belong to 8  species at Ningbo ports.Bioinformatics software Mega 5.0 was applied for phylogeneticanalysis.  Results   The sample of DXR1110 shared highestsequence homology with Mus musculus and the that of DXR1203 sharedhighest sequence homology with Rattus norvegicus. Phylogenetic treeshow different kinds of rat species forming monophyletic grouprespectively.   Conclusion   Analysis of Cytb genesequences is an effective method to indentify rat species.
