ERIC-PCR用于珠海口岸霍乱弧菌的分子分型研究

  摘要:目的   针对珠海口岸分离的霍乱弧菌进行分子分型研究。方法  选择46株菌株(包括45株霍乱弧菌和1株弧菌属细菌), 采用肠杆菌基因间重复一致序列-聚合酶链反应技术(EnterobacteialRepetitive Intergenic Consensus-polymerase chain reaction,ERIC-PCR)扩增细菌的基因组DNA, 对产生的指纹图谱进行聚类分析。结果  ERIC-PCR绘制的基因组指纹图条带清晰可辨且有一定的多态性, 聚类分析结果显示ERIC-PCR将46株菌株分为22个聚类群, 其中从珠海口岸供澳水产品中分离的30株O1群霍乱弧菌非流行株被细分成12个聚类群。ERIC-PCR能有效区分O1群、O139群和非O1/非O139群霍乱弧菌血清群,也能完全区分霍乱弧菌流行株与非流行株。结论  ERIC-PCR是一种有效的、具有高识别力的霍乱弧菌分型方法。本研究获得的霍乱弧菌分子分型数据为珠海口岸霍乱的防控提供了有价值的基础资料。

  Study on molecular subtyping of  vibrio choleraeisolated

  at Zhuhai port by ERIC-PCR

  Abstract:   Objective   To study the molecularsubtyping of vibrio cholerae isolated at zhuhai port.  Methods   The genomic DNA of forty-six strains (includingforty-five vibrio cholerae of different serogroups and one vibriospp.) were amplified by using enterobacteial repetitive intergenicconsensus-polymerase chain reaction (ERIC-PCR). The generatedfingerprint was studied by clustering analysis.   Results  The fingerprinting pattern of ERIC-PCR was clearlydistinguishable with certain polymorphisms. The results of clusteranalysis showed that forty-six strains can be divided intotwenty-two groups by ERIC-PCR. Thirty non-epidemic strains of O1group V. cholerae isolated from seafood which will be exported toMacau at zhuhai port were subdivided into twelve groups. ERIC-PCRwas useful to categorize O1, O139 and non-O1/non-O139 V. choleraeserogroups. Epidemigenic strains and non-epidemigenic strains of V.cholera can also be discriminate completely by it.  Conclusion   ERIC-PCR is an effective approach for subtypingof V. cholerae with high discriminative power. The data ofmolecular subtyping of V. cholerae obtained in this study offeredvaluable basic information for prevention and control of cholera atzhuhai port.
